fa شناسایی گونه‌های مالاسزیا در بیماران مبتلا به درماتیت سبورئیک با استفاده از روش PCR-RFLP عمومى General پژوهشي Research <div style="direction: rtl"><strong style="line-height: 2.2">زمینه و هدف:</strong> مالاسزیا قارچی دوشکلی و چربی‌دوست است که دارای گونه‌های مختلفی می‌باشد. بعضی از آن‌ها به صورت فلور طبیعی روی پوست وجود دارند و در شرایطی ایجاد بیماری می‌کنند. هدف اصلی از این مطالعه، بررسی گونه‌های مالاسزیا در موارد درماتیت سبورئیک با استفاده از روش مولکولی PCR-RFLP بود.</div> <div style="direction: rtl"><strong style="font-weight: bold line-height: 2.2">روش اجرا:</strong> از در این مطالعه‌ی توصیفی، 70 نمونه‌ی جداشده از ضایعه‌های پوستی مبتلایان به درماتیت سبورئیک مورد مطالعه قرار گرفت. شناسایی ایزوله‌ها با استفاده از روش مولکولی PCR-RFLP انجام شد. در این روش ناحیه‌ی (Internal Transcribed Spacer 2(ITS2 توسط پرایمرهای ITS3 و ITS4 تکثیر گردید و توسط سه اندونوکلئاز تحدیدی AluI، BanI و MspAI قطعات مشخصی برای هر گونه‌ی مالاسزیا به‌دست آمد.</div><div style="direction: rtl"><strong style="font-weight: bold line-height: 2.2">یافته‌ها: </strong>گونه‌های جداشده از بیماران به ترتیب مالاسزیا گلوبوزا (48.6%)، مالاسزیا فورفور (40.0%)، مالاسزیا اسلوفیئی (8.6%) و مالاسزیا سیمپودیالیس (2.8%) بودند. هیچ موردی از M. pachydermatis، M. japonica، M. dermatis، M. restricta، M. obtusa، M. nana و M. yamatoensis از نمونه‌ها جدا نشد.</div><div style="direction: rtl"><strong style="font-weight: bold line-height: 2.2">نتیجه‌گیری: </strong>شایع‌ترین گونه‌ی ایزوله‌شده از ضایعات درماتیت سبورئیک، مالاسزیا گلوبوزا و پس از آن مالاسزیا فورفور بود.</div> <p style="text-align: justify"><strong>Background and Aim:</strong> Malassezia is a lipophilic and dimorphic fungus which has different species. Some of them can be found as natural flora on the skin and in some conditions may cause seborrheic dermatitis. The aim of this study was to identify Malassezia species associated with seborrheic dermatitis in Iranian patients, using PCR-RFLP.<br><strong>Methods:</strong> In this study out of 79 patients with seborrheic dermatitis, isolates of 70 patients were positive for Malassezia species using PCR-RFLP. The Internal Transcribed Spacer 2 (ITS2) region was amplified by PCR employing the ITS3 and ITS4 primers and The restriction endonucleases AluI, BanI and MspAI were selected for producing distinct RFLP patterns.<br><strong>Results: </strong>M. globosa (48.6%), M. furfur (40.0%), M. slooffiae (8.6%) and M. sympodialis (2.8%), were the microorganisms responsible for the infection among participants.
