fa بررسی پلی‌مورفیسم کدون 72 ژن p53 در نمونه‌های اسکواموس سل کارسینومای دهانی و مقایسه آن با افراد سالم به روش PCR عمومی general تحقیقی Research <p></p><p dir="rtl"><strong>زمینه وهدف:</strong> پلی‌مورفیسم تک نوکلیوتیدی در کدون 72 ژنp53، ساختار پروتیینp53 را تغییر و فعالیت آن را تحت تاثیر قرار می‌دهد .با توجه به اینکه پلی‌مورفیسم  مزبور وابسته به موقعیت جغرافیایی و نژادی است و فراوانی برخی از سرطا‌ن‌های انسانی را با آن مرتبط دانسته‌اند، هدف از مطالعه حاضر ارزیابی این پلی‌مورفیسم در نمونه‌های SCC دهان ومقایسه آن با افراد سالم در شهر اصفهان بود.</p><p dir="rtl"><strong>روش بررسی:</strong> در این مطالعه case -control 20 بلوک پارافینی با تشخیص  SCCغیر متاستاتیک با تمایز خوب حفره دهان از آرشیو بخش آسیب شناسی دهان دانشکده دندانپزشکی در بین سال‌های 1380-1384 و نیز 20 نمونه خون افراد سالم به عنوان شاهد ،با تکنیک PCR ،مورد مطالعه قرار گرفت. پس از استخراج DNA، با استفاده از پرایمرهای اختصاصی آرژینین و پرولین و نیز الکتروفورز، ژل آگارز پلی‌مورفیسم کدون 72 ژن p53 مشخص گردید. اطلاعات بدست آمده با استفاده از نرم‌‌افزار SPSS مورد تجزیه و تحلیل آماری قرار گرفت. برای مقایسه توزیع فراوانی سه ژنوتیپ کدون 72 از آزمون دقیق فیشر با سطح معنی‌د‌اری 05/0 >pاستفاده شد.</p><p dir="rtl"><strong>یافته‌ها: </strong>شیوع ژنوتیپ آرژینین/ آرژینین، آرژینین/ پرولین و پرولین / پرولین در بیماران به ترتیب 45%، 45% و 10% و در گروه کنترل 45%،50% و 5% بود که اختلاف آماری معنی‌داری را نشان نداد.</p><p dir="rtl"><strong>نتیجه‌گیری:</strong> ارتباط بین پلی‌مورفیسم کدون 72 ژن p53 و بعضی سرطان‌ها در مناطق مختلف مشاهده شده است. با این وجود بر اساس نتایج مطالعه حاضر، این پلی‌مورفیسم را نمی‌توان به عنوان عامل مستعد کننده ژنتیکی در ابتلاء به SCC دهان در شهر اصفهان  منظور نمود.</p></span /> <p style="text-align: justify"><strong>Background and Aim:</strong> A single nucleotide polymorphism at codon 72 of the p53 gene alters the p53 protein structure and affects its activity. This polymorphism depends on geographic regions and race. Also its association with some cancers has been reported. The aim of this study was to investigate this polymorphism in well differentiated oral SCC and normal population in the city of Isfahan.</p> <p style="text-align: justify"><strong>Materials and Methods:</strong> In this case-control study, 20 paraffin blocks of non metastatic and well differentiated oral SCC were selected from the archive of oral pathology department of dental school between 2001 and 2005. 20 whole blood samples from normal people were considered as control group. After DNA extraction, p53 codon 72 polymorphism was determined by polymerase chain reaction (PCR) technique using specific primers of Arg and Pro and agarose gel electrophoresis. Data were analyzed by Fisher's exact test with p<0.05 as the level of significance.</p> <p style="text-align: justify"><strong>Results:</strong> The prevalence of Arg/Arg , Arg/Pro and Pro/Pro genotypes in case group were 45%,45% and 10% respectively compared to 45%,50% and 5% in controls.
