other بررسی بیان ژن سه گیرنده آدرنرژیک آلفا 1، 2 و بتا 2 سلول‌های کومولوس تخمدان زنان نابارور با پاسخ ضعیف تخمدانی کاندید لقاح آزمایشگاهی Evaluation of gene expression of three adrenergic receptors in infertile women with poor ovarian response, candidate for IVF مقاله اصیل Original Article <div style="text-align: justify;">زمینه و هدف: بیشتر مطالعات نشان می‌دهد ۹ تا ۲۴% افرادی که در سیکل لقاح آزمایشگاهی (IVF) قرار می‌گیرند زنانی هستند که پاسخ تخمدانی آن‌ها ضعیف است. زنان با پاسخ ضعیف تخمدانی Poor ovarian response (POR) بیماران ناباروری هستند که ذخیره تخمدان و پاسخ تخمدان به داروها کم و کیفیت تخمک در آن‌ها رو به زوال است. هدف این مطالعه بررسی بیان ژن سه گیرنده آدرنرژیکی در سلول‌های کومولوس زنان پاسخ ضعیف تخمدانی در محیط کشت بود. روش بررسی: این مطالعه موردی-شاهدی از فروردین تا دی ۱۳۹۶ در مرکز تحقیقات بهداشت باروری بیمارستان امام خمینی (ره) تهران بر روی دو گروه مطالعه و کنترل (اهدا کننده تخمک) انجام شد. هر دو گروه با ضوابط محدوده سنی ۲۰ تا ۴۵ سال و داشتن اندکس توده بدنی زیر m2/kg ۲۸ وارد مطالعه شدند و ضوابط خروج شامل عدم مصرف دارو بجز داروهای تحریک‌کننده تخمدان و عدم ابتلا به بیماری بود. تشخیص پاسخ ضعیف تخمدان با معیار بولونیا (Bologna) انجام شد. پس از پانکچر فولیکول‌ها، کومولوس‌ها از کمپلکس تخمک-کومولوس جدا و با لام نئوبار شمارش و به محیط کشت اضافه شد. استخراج RNA انجام و غلظت RNA خوانده شد. cDNA سنتز و برای ژن‌های B2 و ADRα1,2 پرایمر طراحی و بیان ژن سنجیده شد.  یافته‌ها: مقایسه نتایج بیان ژن فقط در گیرندۀ آلفا ۱ و ۲ سلول‌های کومولوس در دو گروه مطالعه و کنترل، کاهش معناداری نشان داد. ضریب همبستگی نیز نشان داد که بین گیرنده‌های آدرنرژیک و اثرات آن‌ها بر یکدیگر رابطه معنادار وجود دارد.  نتیجه‌گیری: نتایج مطالعۀ حاضر کاهش بیان ژن دو گیرنده آلفا ۱ و ۲ در این زنان را نشان داد.</div> <div data-select-like-a-boss="1" style="text-align: justify;">Background: Most studies show that 9 to 24% of people who are in in vitro fertilization (IVF) cycles are women who respond poorly to ovarian stimulation. Women with poor ovarian response (POR) are a group of infertile patients whose ovarian reserve, ovarian response to medication, and the quality of ovum are declining. Therefore, the number of female cycles, the number of fetuses from the oocyte and the rate of pregnancy in these women is reduced. The purpose of this study was to investigate the expression of three adrenoceptor receptor genes in the cumulus cells of women with poor ovarian response in culture medium. Methods: This case-control study was conducted in two groups: study (POR) and control (oocyte donor's women) groups. POR diagnosis was performed by ESHRE Bologna criteria. After puncture of the follicles, cumulus-oocyte complex was collected and the cumulus cells (CCs) were isolated by enzyme and are counted with Neobar lamella and then were added in the culture medium. After completing the culture, RNA was extracted from cumulus cells and the RNA concentration was read by the Spectrophotometer (NanoDrop Technologies, Wilmington, DE, USA). Then cDNA synthesized and primers designed for ADR-α1, 2 and ADR-B2 or gene expression by real-time polymerase chain reaction (PCR) technique. The research was done in Reproductive Health Research Center of Imam Khomeini Hospital, Tehran University of Medical Sciences, Tehran, Iran, from April to December 2017. Results: Comparison of the results of ADR-α1, 2 gene expressions in cumulus cells showed a significant decrease, but ADR-B2 was not significant in two groups. Correlation coefficients also showed that there are relationship between three adrenoceptors and their effects on each other.
