Abstract
Background: Leptin, a peptide hormone, is the product of "ob" Gene. Leptin regulate body weight and composition through reducing appetite and energy expenditure in rodents and humans. The aim of this study was to evaluate differences in expression of Leptin Gene in different tissues of streptozotocin induced diabetic rats.
Methods: 40 Sprague Dawely rat were selected. Intra peritoneal injection was carried out in 20 rats and another 20 rats were used as control. After injection of 60mg/kg Streptozotocin, animals were transformed into diabetic. Glucose was measured by glucose oxidase method. Leptin and insulin were measure by commercially available immunoassay kits. After one week treatment, different tissues including adipose tissues, Spleen, epidydimis, and Liver of both control and experimental animals were dissected. For investigation of any changes of the Leptin gene expression in different tissues, RNA was extracted using Trizo1 method. By using RT-PCR technique, Leptin cDNA and β-actin cDNA as internal control were constructed and PCR was carried out. The RT-PCR products were detected on 2% agarose gel using electrophoresis.
Results: Mean serum levels of Leptin was 5.23± 0.45 ng/ml before injection of streptozotocin and markedly decreased in STZ induced diabetic rats to 0.79±0.25 ng/ml. This decrease was statistically significant P<0.05). There was a direct and significant correlation between leptin and insulin in streptozotocin-induced diabetic rats (r=0.37, P<0.05 ) while, this was reverse in control rats ( r= -0.28, P<0.05). Using RT-PCR method, Leptin gene expression in different tissues including fat epidydimis, liver, and spleen showed that the intensity of leptin band with 452 bp was decreased in diabetic rats in comparison to normal rats. Actin Gene expression was identified in PCR products having 403 bp and the intensity was constant in both groups. The reduction rates of "ob" mRNA in fat epidydimis tissue in STZ diabetic rats was remarkable in comparison to Spleen and Liver.
Conclusion: It is speculated that Leptin gene could be under regulation of insulin dependent mechanism in diabetic rats and by modulating Leptin gene expression in diabetic patients, it may be useful in clinical practices.

Abstract
Background: Diabetes is a common endocrine disease in human kind. In most type II diabetic patients, obesity and overweight status account as the serious health problems worldwide and variety of endocrine factors well known that have regulatory role in weight balance and body composition including Leptin and IGF-I factor. The aim of this study was to examine the correlation between Leptin and IGF-I in type II diabetics and controls.
Methods: As a case- control study, 38 type 2 diabetics (20 males and 18 female with mean age 49.22) and 46 healthy controls (16 males and 30 females with mean age 49.52) are recruited. We measured the concentrations of FPG, IGF-I, HbA1C and IGFBP-3 in both groups. FPG was measured by enzymatic glucose oxidase method and Hb Gold analyzer HPLC was used to measure HbA1C. Determination of Leptin, IGF-I, IGFBP-3 and Insulin concentrations were carried out using ELIZA method. P< 0.05 was considered as statistically significant.
Results: The mean of BMI and age were not significantly different in both groups. The mean serum levels of IGF-I, Leptin, Insulin, FPG and HbA1c concentrations in type II diabetics were significantly higher than controls (P< 0.05). In males, the mean serum levels of Leptin were statistically lower than in females in both groups. There was a strong correlation between IGF-I and IGFBP-3, Leptin and insulin, IGF-I and age, and BMI with FPG in both patients and controls (P< 0.05). A reverse correlation was observed between IGF-I and HbA1c in patients and controls (P< 0.05).
Conclusion: It is speculated that based on this findings, Leptin and IGF-I system could have regulatory roles in body composition and fat content particularly in obese and overweight diabetic patients and have significant correlation with Insulin, glucose, BMI and age.

Abstract
Background: Obesity is an escalating public health problem. It is a major risk factor for atherosclerosis, hypertension, and type 2 diabetes. Since circulating levels of the adipocytokins are associated with obesity and dyslipidemia, we investigated the relationship of plasma adipocytokine concentrations with VLDL apolipoprotein B (apoB)-100 kinetics in men.
Methods: Plasma adiponectin, leptin, resistin, interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) concentrations were measured using enzyme immunoassays and insulin resistance by homeostasis model assessment (HOMA) score in 41 men with BMI of 22–35 kg/m2. VLDL apoB kinetics were determined using an intravenous infusion of 1-[13C]leucine, gas chromatography–mass spectrometry, and compartmental modeling. Visceral and subcutaneous adipose tissue mass (ATM) were determined using magnetic resonance imaging, and total ATM was measured by bioelectrical impedance.
Results: In univariate regression, plasma adiponectin and leptin concentrations were inversely and directly associated, respectively, with plasma triglyceride and HOMA score. Conversely, adiponectin and leptin were directly and inversely correlated, respectively, with VLDL apoB catabolism and HDL cholesterol concentration (P < 0.05). Resistin, IL-6, and TNF-α were not significantly associated with any of these variables. In multivariate regression, adiponectin was the most significant predictor of VLDL apoB catabolism (P= 0.001) and, together with visceral ATM, was an independent predictor of plasma VLDL apoB concentration (P = 0.015) HOMA score was the most significant predictor of VLDL apoB hepatic secretion (P= 0.049). Leptin was not an independent predictor of VLDL apoB kinetics.
Conclusion: Plasma VLDL apoB kinetics may be differentially controlled by adiponectin and insulin resistance, with adiponectin regulating catabolism and insulin resistance regulating hepatic secretion in men. But leptin, resistin, IL-6, and TNF-α do not have a significant effect in regulating apoB kinetics.

Abstract
Background: Type 2 diabetes is a major metabolic disorder. This study was aimed to evaluate the effect of probiotic soymilk fermented with Bifidobacterium lactis and containing omega-3 on hematologic, oxidative stress, anti-oxidant and inflammatory parameters in type 2 diabetic rats.
Methods: In this experimental manipulation study 65 rats were divided into five groups. Type 2 diabetes was induced by a single injection of streptozotocin and nicotinamide. Normal control (NC) and diabetic control (DC) rats received 1 mL/day of distilled water, and three groups of diabetic rats were administered 1 mL/day of soymilk (SM), fermented soymilk (FSM) and fermented soymilk fortified with omega-3 (FSM+omega-3) products by oral gavage for 28 days. Hematologic, oxidative stress, anti-oxidant and inflammatory parameters were measured at the end of experiment.
Results: At the end of experiment, in all treated diabetic rats compared with the DC group, red blood cell (RBC), hemoglobin (Hb), hematocrit and superoxide dismutase (SOD) concentrations increased significantly (p<0.05), and malondialdehyde (MDA) concentrations reduced significantly (p<0.05). The reduction of MDA and increasing in Hb, RBC, hematocrit and SOD concentrations in the FSM+omega-3 group was more than two other groups. In the FSM+omega-3 group, high sensitivity C-reactive protein (hs-CRP) concentrations compared with the DC and FSM groups were significantly lower (p<0.05).
Conclusion: The experiments suggest that combining soymilk, probiotics and omega-3 may be beneficial for the reducing oxidative-stress in type 2 diabetes.

Abstract
Background: Neuropathic pain is associated with a profound impact on quality of life and daily activity in patients and caused high medical costs. Because of the nerves destructive nature in this disease, the neurotrophins are high regarded for their possible effects on survival, growth and development and neuronal functions as a possible therapeutic strategy. So the aim of this study is investigation of the chronic effects of decreased activity in neuropathic pain form on the NFG gene expression in the sciatic nerve of Spinal nerve ligated rats.
Methods: Ten adult male Wistar rats in the weight range of 250±30 gr randomly were divided into two groups including healthy control (C), ligation sciatic nerve ligation. Over the six weeks neuropathic pain behavior tests were conducted continually in groups. In the end change of NGF gene expression in sciatic nerve was measured with Real time PCR technique and calculated using the 2-&Delta&DeltaCT method.
Results: After 6 weeks, neuropathic pain behavior tests showed that pain threshold of thermal hyperalgesia and mechanical allodynia in the SNL group was significantly lower than the control group (P&le0.05). In addition, NGF gene expression in sciatic nerve ligation group compared to controls increased significantly (P&le0.05).
Conclusion: The findings of this study suggest that the development of neuropathic pain and decreased physical activity is associated with increased endogenous NGF. Although it is not clear that increase in NGF expression is due to the nature of neuropathic pain or nerve damage, it is clear that excessive expression of NGF is associated with neuropathic pain.

Abstract
Background: Diabetic Nephropathy is one of the main microvascular complications of diabetic mellitus. Methylenetetrahydrofolate Reductase (MTHFR) is one of the candidate genes of diabetic nephropathy. MTHFR (C677T) polymorphism reduces catalytic activity of MTHFR and leads to increase level of plasma homocysteine. The aim of this study was to evaluate the association of C677T polymorphism with diabetic nephropathy.
Methods: In this case control study, 300 individuals, including type 2 diabetes mellitus with diabetic nephropathy (N=104), diabetes mellitus patients without diabetic nephropathy (N=100) and controls (N=96) participated. The MTHFR genotype was determined using PCR-RFLP technique and biochemical parameters were measured.
Results: Genotype frequencies were significantly different between patients with diabetic nephropathy and diabetes mellitus without nephropathy (TT+CT vs CC; P=0.02,OR:0.5,CI:0.3-0.9).The allele frequency was also significantly different between diabetic nephropathy and diabetics mellitus without nephropathy(P=0.013,OR:1.754,CI:1.123-2.740).
Conclusion: These findings suggest that there is an association between C677T polymorphism and nephropathy in patients with type 2 diabetes. Allele C increase the risk of nephropathy, and T allele has a protective role in susceptibility to disease.

Abstract
Background: Zinc-Alpha 2-Glycoprotein (ZAG) has recently been implicated in the regulation of adipose tissue metabolism due to its negative association with obesity and insulin resistance. The purpose of this study is to investigate the effect of eight weeks HIIT on level of ZAG in plasma and adipose tissue in type 2 diabetic male rats.
Methods: Twenty one male rats were divided into the three groups of sham control (healthy), control (diabetic), and interval training (diabetic with training). The training group received 8 weeks of training sessions each with 5-12 repetitions of high intensity training for 15-30 seconds at the speed of 27-34 meters per second on a treadmill followed by one minute of active rest. Twenty four hours after the training session blood and body fat samples were taken to measure ZAG levels. The data was analyzed using one-way ANOVA and Tukey’s post hock test at the significance level of 0.05 (P<0.05).
Results: There were significant differences between the sham control and the control with respect to ZAG contents in adipose tissue, which was lower in the diabetic control group (P<0.001). Moreover, significant differences in ZAG contents of adipose tissue were also observed between the sham control and the group receiving the training, with the sham control having higher ZAG contents in adipose tissue (P<0.005). The eight-week HIIT significantly increased the amount of secreted ZAG in adipose tissue compared to the control diabetic group (P=0.003). No significant differences were recorded between the groups in ZAG plasma levels.
Conclusion: The HIIT increased ZAG content in the adipose tissue of the male diabetic rats. This can reduce insulin resistance in type 2 diabetes.

Abstract
Background: The main problem of modern humans is non-communicable diseases such as diabetes, cardiovascular disease, etc. Diabetes has the highest prevalence. It imposes many costs on society and the patient. Diabetics have special needs for a better life; hence, the researcher's aim is to accomplish them by defining researches and technology missions. Our purpose was to understand the real needs of a diabetic patient. Previous studies were evaluated to find out which one was effective.
Methods: In this research, 120 diabetic children's parents were requested to write a letter entitled "Hi Inventor" and describe any efficient inventions, which could make life easier for their children. Besides, research papers and patents from Iran over the past ten years have been evaluated in terms of their compliance with patients' needs.
Results: Parent's statements gathered in the form of 12 real needs. The main concern was the invasive method of blood glucose testing and they were asking for innovative method to measure blood glucose with less pain. Among published articles by Iranian researchers over the past ten years, 3% of the articles were in some way related to the actual needs of patients with diabetes. A total of 1017 patents from Iran was looked for, there were 3 diabetes-related patents.
Discussion: To sufficiently support the conducting research and identifying research priorities in the country, it is necessary to consider a balanced ratio between fundamental and applied research so that the patients' community can benefit from the early results of research as well.

Abstract
Background: Type 2 diabetes mellitus (T2DM) is the most common type of diabetes that was classically characterized by pancreatic β-cell dysfunction. Changes in circadian patterns is one of the reasons which can increase the occurrence of diabetes. Melatonin is one of the biological molecules which plays an important role in regulating the circadian clock and also an inhibitory effect on insulin secretion in β-cells. The aim of this study was to examine the association between MTNR1B (rs10830962) gene polymorphism and the risk of T2DM.
Methods: Genotyping was carried out in a total number of 208 subjects including 108 patients with T2DM and 100 normal controls using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) which is confirmed by Sanger sequencing method.
Results: The frequencies of CC, GC and GG among cases were 54.63%, 1.85% and 43.52% and in control subjects were 81%, 0% and 19% respectively (P<0.001). Frequency of G allele among diabetic patients was significantly higher than non-diabetics (OR=3.34, CI=2.10-5.36, P<0.001).
Conclusion: Our study showed that rs10830962 polymorphism of the MTNR1B gene can be directly associated with T2DM risk.

Abstract
Background: Type 2 diabetes Mellitus (T2DM) is a multifactorial, polygenic disease caused by impaired insulin secretion, insulin resistance and beta-cell dysfunction. Melatonin is a circadian rhythm regulator and any imbalance in its levels can be related to various metabolic disorders. Melatonin and the genetic variants of MTNR1B gene are reported to be associated with T2DM susceptibility. We investigated the association between rs4753426 variant in the MTNR1B gene and the risk of T2DM in group of Iranian patients.
Methods: In this case-control study108 T2DM and 100 normal individuals were recruited to genotyping by PCR- RFLP.
Results: It was observed a significant difference in CC, CT, and TT genotypes distribution between T2DM and control groups (P<0.001). Frequency of C allele among cases was significantly lower than controls (8.3% vs. 42.5% respectively, P<0.001) and C allele carriers had a 88% lower risk of developing T2DM than T carriers.
Conclusion: Our results showed that the rs4753426 variant of MTNR1B gene could reduce the risk of T2DM developing.

Abstract
Background: Diabetes causes nerve tissue damage and ultimately death of nerve cells, especially in important areas such as the hippocampus. It seems that exercise training and anti-inflammatory supplements can moderate this condition. Therefore, the aim of this study was to determine the effect of aerobic training exercise and L-carnitine consumption on the levels of hippocampus CNTF and CNTFR proteins in diabetic rat.
Methods: 45 Wistar rats between 250 to 300 gr were randomly divided into six groups, including healthy control groups, sham, diabetic control, diabetes
+ aerobic exercise, diabetes + L-carnitine supplementation and diabetes + aerobic exercise + L-carnitine supplementation. L-carnitine supplements were divided. Diabetic groups underwent subcutaneous injection of 55 mg / kg STZ. supplementary groups received 100 mg of L-carnitine per day orally. The aerobic exercise program was performed for 6 weeks and 5 days a week. The research variables were measured 24 hours after the last training session by the ELISA technics in the hippocampal tissue. The statistical method of one-way analysis of variance and Toki post –hoc test were used to analyze the data.
Results: The present study showed an significant increase in CNTF and CNTFR protein levels of hippocampal tissue after aerobic exercise and L-carnitine consumption (P = 0.001), which greater effect found in combination manner.
Conclusion: Due to the effect of aerobic exercise and L-carnitine supplementation alone and in interaction with each other on hippocampal tissue, the use of this method can be considered as maintaining neural tissue in diabetes.

Abstract
Background: In this study, we investigated the diagnostic power of ultrasound in the diagnosis of malignancy in thyroid nodules in Iranian patients. For this purpose, we examined the relationship between ultrasound findings and pathology findings.
Methods: The present study is a retrospective study. The patients with a diagnosis of thyroid nodules who underwent ultrasound and FNA, were included in this study. To assess the relationship between the results of FNA and the ultrasound characteristics of nodules, we classified the results of FNA into malignant and benign groups and then compared ultrasound characteristics between the two groups. In cases which the FNA results were indeterminate (AUS/FLUS or FN/SFN), the postoperative pathology result was considered (if thyroid surgery was done and the result was available).
Results: In total, 201 nodules were included in this study. The results showed that hypoechogenicity, irregular/ill-defined margin, microcalcification, malignant flow pattern in Doppler sonography and concurrent cervical lymphadenopathy with suspicious features were significantly associated with malignant thyroid nodules. However, other ultrasound findings, such as the size and location of the nodule, presence of a cystic components within the nodule, the presence of a Halo sign, and the presence of a taller-than-wide shape, could not distinguish between benign and malignant nodules. Finally, the results of the present study showed that the accuracy of ultrasound in the diagnosis of malignancy in thyroid nodules is high.
Conclusion: This study suggests that the use of ultrasound can be very effective in diagnosing malignancy in thyroid nodules.

Abstract
Background: The release of adipokines from adipose tissue depots plays a key role in regulating metabolic homeostasis and several other physiological processes, including diabetes, obesity, and vascular diseases. This study investigated the effect of eight weeks of high intensity interval training (HIIT) on asprosin, lipid profile and insulin resistance in type 2 diabetic male rats.
Methods: 24 male Sprague Dawley rats were randomly divided into four equal groups: control (C), control traning (C+T), diabet (D) and diabet traning (D+T). Diabetes was induced by the combined method of high fat diet and low dose strepotozocin injection. The traning group performed the HIIT program on the treadmill for eight weeks. Data were analyzed using one-way ANOVA and bonferroni post hoc test at a significance level of P<0.05.
Results: The results showed increased plasma asprosin in D group compared to C (P=0.0001) and decreased in C+T group compared to C (P=0.03) and D+T group compared to D (P=0.04). There was no significant difference in HOMA-IR between the C and C+T group (P=0.9) but decreased in D+T compared to D (P=0.0001). HDL increased in D+T group compared to the D (P=0.0001) and decreased TG and LDL (P=0.001). There was no significant difference between TG and LDL in the C group compared to the C+T, but HDL increased in C+T (P=0.01).
Conclusion: Plasma asprosin increases in rats with type 2 diabetes and HIIT can reduce the complications of diabetes by improved lipid profile and reduce asprosin and insulin resistance.

Abstract
Background: The effects of regular exercise on miRNA profile and oxidative stress indices are less known. The aim of this study was to investigate the changes in expression of miR-17 and oxidative stress indices in overweight and obese women following high-intensity functional training.
Methods: In this semi-experimental study, 22 overweight and obese women were randomly divided into two groups of high-intensity functional training (n= 10) and control (n= 12). The subjects of the training group performed high-intensity functional exercises 3 times a week for 8 weeks and intensity of rating of perceived exertion≥7. Before and after 8 weeks, blood samples were taken to measure expression of miR-17 gene, malondialdehyde (MDA), superoxide dismutase, glutathione peroxidase (GPx) and total antioxidant capacity (TAC). Analysis of covariance test was used to analyze the data.
Results: After eight weeks of high-intensity functional training, expression of miR-17 in the experimental group significantly decreased compared to the control group (P= 0.001). After eight weeks of high-intensity functional training, TAC (P= 0.0001), GPX (P= 0.007) and SOD (P= 0.001) increased significantly in the experimental group compared to the control group. No significant change was observed in MDA (P= 0.778).
Conclusion: The results of the present study showed that eight weeks of high-intensity functional training may lead to the reduction of miR-17 expression and the improvement of oxidative stress indices and antioxidant enzymes, which may be effective in improving metabolic indices and obesity complications.

Objective: Obesity is known to have metabolic complications including insulin resistance (IR) and metabolic disorder. The aim of this study was to compare the effect of two traditional 12-week circular resistance training methods on the levels of branched and aromatic amino acids and insulin resistance of overweight and obese men.
Design: Thirty-three overweight and obese men with mean and standard deviation of age, weight and body mass index were 17.7±0.13 years, 92±2.45 kg and 30.70±32 1.32 kg / m2, respectively. They were selected and randomly divided into three groups: circular resistance training (n = 11), traditional resistance training (n = 11) and control (n = 11). The training program consisted of two traditional resistance training protocols and 12-week wave circuits that were exactly the same volume; Which was run three times a week. Blood sampling was performed 48 hours before and after the training protocol and after 8 hours of fasting. HPLC was used to measure serum levels of amino acids.
Results: The results of analysis of variance in the groups showed that circuit training more than 2 times compared to traditional training reduces total BCAAs (35.9 vs. 15.5%) and AAAs in serum (19.5 vs. 9%) and equally the level of insulin resistance. (23 and 26.7 percent, respectively) reduced the door in overweight and obese young men.
Conclusion: The results of this study showed that the level of changes in circular resistance training was higher than traditional. Circular resistance training may be able to prevent obesity-related metabolic disorders.

Background: Pharmacogenomics (PGx), as a growing field of personalized medicine, aims to optimize the efficacy and safety of medications by studying the association between germline genetic variations and drug responses. The present cross-sectional study aims to evaluate the allele frequency of the NUDT15 genetic variant in the Iranian population to provide insights into personalized treatment decisions in the Iranian population.
Methods: A representative sample set of 1142 unrelated healthy Iranian individuals aged 18 and older genotyped using the Infinium Global Screening Array-24 BeadChip.
Results: We identified a pharmacogenetic variant with minor allele frequency (MAF) ≥1% among the present studied population which may explain the substantial variability in drug response phenotypes among different populations
Conclusion: The results of our study reveal significant genetic variation among Iranian populations that could significantly influence clinical decision-making. This study shows the frequency pattern of the effective variant of NUDT15 in determining the phenotype in Iranian population.
