Abstract

Background: All contemporary methods of insulin administration are non-physiological. Insulin is not absorbed from the gastrointestinal tract because of its peptide nature. The aim of the present study was to examine the absorption of oral insulin from gastrointestinal tract, using novel oral formulation- adding a delivery agent superporouse hydrogel (SPH) and SPH composite (SPHC) in combination with insulin.
Methods: Capsules containing insulin and SPH &SPHC were administered orally, to 15 non-diabetic subjects in order to assess its biological effects and safety. Plasma glucose, insulin and c – peptide serum levels were determined, at timed intervals up to 4 h.
Results: In the present study, we showed that AUC of exogenous insulin in polymer -insulin group was higher than sub-cutaneous regular insulin group. It means that addition of SPHC polymer caused increase in insulin absorbtion.In addition, Insulin Tmax in polymer group was longer than sub-cotaneaus insulin group. Blood glucose AUC in sub-cotaneaus group was higher than polymer group.AUC C-peptide serum level in polymer group was higher than sub-cutaneous group.
Conclusions: Insulin in combination with a novel delivery agent, SPH and SPHC, given orally is absorbed through the GI tract in a biologically active form. This was demonstrated by suppression of endogenous insulin secretion.

Abstract

Background: All contemporary methods of insulin administration are non-physiological. The euglycemia that is achieved in at the expense of the adverse effects of systemic hyper-insulinemia, emphasize the importance of devising methods to deliver insulin physiologically and directly into the portal circulation. The aim of the present study was to evaluate the oral absorption of insulin from gastrointestinal tract, using novel oral drug delivery system delivery based on superporouse hydrogel (SPH) and SPH composite (SPHC) in combination with insulin.
Methods: This study has been done based on interventional clinical trial in healthy volunteers. Capsules containing insulin and SPH &SPHC in various combination were administered orally, to 15 non-diabetics subjects in order to assess this biological effects and safety. Serum glucose, insulin and C - peptide levels were determined, at predetermined timed intervals up to 4 h.
Results: An increase in serum insulin level was demonstrated in all subjects that used polymer plus insulin. The nadir of serum glucose level appeared after 60 - 120 min following the ingestion of polymer plus insulin. Serum C - peptide levels were suppressed while exogenous insulin was absorbed at the same time. No adverse effects were detected during the trial and several weeks following the trial using SPH based drug delivery system.
Conclusions:
Insulin in combination with novel delivery agents, SPH & SPHC, given orally was partially absorbed through the GI tract in a biologically active form. This was demonstrated by serum glucose lowering effect of the delivery system as well as a suppression of plasma C-peptide which also represented a decrease in endogenous insulin secretion.

Abstract

Background: Diabetic foot wound considered as cause of disability in diabetic's patients. And despite the progress made in the diagnosis and treatment of diabetes, since diabetic foot problem has not been resolved. The aim of this study was to investigate the effect of endurance training and cold plasma on wound healing in diabetic rats.
Methods: 25 male Wistar rats were divided randomly into 5 groups (n=5). The once injected intraperitoneally with a concentration of 40mg/kg of body weight was taken. 4mm punch biopsy wounds were used to create skin wound. The exercise protocol, consists of 60minutes of running on a treadmill at a speed of 25m/min, 5days a week for 4times in a week. The whole process of plasma irradiation on wound took over 8times at each of the plasma samples for 10 minutes under direct radiation at a distance of 20 mm from the tip, respectively. The wound picture was taken with the camera Dino-Lite. Given the normal distribution of data to test hypotheses of parametric tests one-way ANOVA and repeated measure ANOVA were used to determine differences (P≤ 0.05).
Results: The findings showed that the difference between exercise+diabetes group and Ctrl+diabetes group wasn’t significant. In addition, the difference between plasma+diabetes group and Ctrl+diabetes group wasn’t significant. However, the significant difference was between plasma+training+diabetes group and Ctrl+diabetes group.
Conclusion: This is the first time that the combination of exercise and cold plasma used for wound healing in diabetes and have significantly accelerated wound healing in diabetic mice.

Abstract

Background: Diabetes produces free radicals and damages the liver. The aim of this study was to investigate the effects of Rosmarinus Officinalis leaf hydroalcoholic extract on histopathology and enzymes activity of liver in alloxan induced diabetic rats.
Methods: In this study, 50 adult male Wistar rats weighing 200 to 250 grams, were divided randomly into 5 groups of 10; non-diabetic control, diabetic control and 3 experimental groups (diabetic rats treated with the dosage of 50mg/kg, 100mg/kg and 200mg/kg rosemary leaf extract intraperitoneally for 14 days).
Diabetes was induced in rats by intraperitoneal injection of a single dose 120 mg/kg alloxan was done. At the end of the treatment period, blood samples were taken from the left ventricular heart of mice and aspartate aminotransferase (AST / SGOT), alanine aminotransferase (ALT / SGPT) and alkaline phosphatase (ALP) in serum were measured. Immediately liver was removed and histological samples were fixed in 10% formalin and then stained with hematoxylin-eosin technique (H & E).
Results: In diabetic rats, unlike control mice, the liver enzymes (AST, ALT and ALP) increases, because of damage of liver tissue (p<0.01).
The level of liver enzymes (AST, ALT and ALP) in rats treated with doses 50mg/kg, 100mg/kg and 200mg/kg of rosemary leaf extract showed a significant reduction in these enzymes compared to control diabetes (p<0.01).
Conclusion: Histological studies showed reduced inflammation in the liver lobule and the port in the experimental groups. The third experimental group had greatest impact on reducing liver inflammation and space ports showed lobule. Histologically, tissue changes were in line with biochemical changes.
The effects of Rosmarinus Officinalis leaf hydroalcoholic extract because of its high antioxidant properties, reduce free radicals and inflammation of the liver damage caused by diabetes by inhibiting the enzyme is reduced.

Abstract

Background: Type 2 diabetes is a metabolic disease and the most common endocrine disorder. Visfatin is a cytokine with insulin-mimetic effects which is associated with diabetes. The rs61330082 polymorphism is located in the promoter region of visfatin gene and regulates visfatin gene expression. The aim of this study is to investigate the association of rs61330082 polymorphism of the visfatin gene with biochemical and anthropometric variables, visfatin level, diabetes and insulin resistance.
Methods: This case‑control study was performed on 80 patients with type 2 diabetes and 80 controls (persons without diabetes). Biochemical and anthropometric parameters were determined using standard methods. Insulin and visfatin levels were measured by ELISA method. Genotype was determined by polymerase chain reaction-restriction fragment length polymorphism method. The data was analyzed with SPSS software.
Results: LDL-C level in healthy people and fasting blood sugar level in patients showed a significant difference in carriers of rs61330082 polymorphism genotypes. No associations was found between visfatin level and single nucleotide polymorphism rs61330082 in the current study. Regression analysis showed that genotypes of rs61330082 polymorphism are not significantly associated with type 2 diabetes and insulin resistance.
Conclusion: The rs61330082 polymorphism is associated with LDL-C in healthy subjects and FBS in patients with T2D. Visfatin rs61330082 polymorphism is not associated with visfatin level and type 2 diabetes and insulin resistance.

Abstract

Background: The role of the immune system and inflammation in controlling metabolic diseases such as insulin resistance, type 2 diabetes, obesity and overweight is well established. Also, physical activity controls and prevents the occurrence of metabolic diseases, which has anti-inflammatory and anti-oxidative effects. The aim of the present study was to study the effect of eight weeks of 10, 20, 30 seconds exercise training on the levels of antimicrobial factors in the saliva of obese adolescent boys.
Methods: 32 male students voluntarily participated in the present study and were randomly assigned to four groups: 1) obese aerobic exercise, 2) normal weight aerobic exercise, 3) obese control without exercise, and 4) normal weight control without exercise. The criteria for the obese group was a BMI above 25. Anthropometric indices of height, weight and body mass index were measured exactly. Before and after eight weeks of practice, Shatell-Run standard test was performed. Salivary samples of lysozyme, lactoferrin, lactate and C-reactive protein concentrations were taken after eight weeks of training with a frequency of three sessions per week with an intensity of 30 to 90% of maximum aerobic power. Using analysis of covariance, variables with a significance level of less than (P≤0.05) were included in the analysis.
Results: The results showed a significant increase in salivary levels of lactoferrin (P=0.001), lysozyme (P=0.003), lactate (P=0.001), and C-reactive protein (P=0.006). There are more obese students than people of normal weight (Youth body mass index criterion)
Conclusion: The increased response of some antibacterial and inflammatory salivary proteins after eight weeks of 10-20-30 sports training following exhaustive activity may be due to the acute response of the immune system.

Abstract

Background: Alzheimer's disease is the most common type of dementia, which is associated with progressive loss of memory and reaction to the environment. The aim of this study was to investigate the effect of 12 weeks of resistance training with sumac intake on the serum levels of SIR3, SIR4 and AMPK in Alzheimer's male rats.
Methods: In this experimental study, 35 Wistar rats aged 8-10 weeks and average weight of 230±30 grams were randomly divided into five groups: control, Alzheimer's disease, Alzheimer's disease with sumac supplementation, Alzheimer's disease with resistance training, and Alzheimer's disease with They were divided by resistance training and receiving sumac supplements. To induce Alzheimer's disease, 8 mg/kg of trimethyl tin chloride along with 200 microliters of normal saline were injected into mice. Resistance training was performed for 12 weeks and 5 days a week on a special rat ladder. 48 hours after the last training session, a blood sample was taken. Serum levels of SIR3, SIR4 and AMPK were evaluated by ELISA method. Results: the present research showed that 12 weeks of resistance training increases the levels of SIR3 (P<0.036), SIR4 (P<0.029) and AMPK (P<0.047) in Alzheimer's rats. In addition, adding sumac to the resistance training program had no significant effect on SIR3 (P<0.342) and SIR4 (P<0.99).
Conclusion: Our results suggest that resistance training is associated with improving the levels of sirtuin 3, sirtuin 4 and adenosine monophosphate kinase in Alzheimer's rats, and receiving sumac antioxidant compound does not lead to a doubling of these responses

Abstract

Background: Adenosine monophosphate-activated kinase (AMPK) is a key regulator of cellular metabolism, and its dysregulation is associated with metabolic diseases such as obesity, inflammation, diabetes, and cancer. Therefore, the purpose of this research is the effect of moderate intensity interval training (MIIT) on the total and phosphorylated content of AMPKα1/2 protein in the skeletal muscle of diabetic rats.
Methods: In this experimental study, 12 two-month-old male Sprague Dawley rats with an average weight of 280±30 grams were selected. Diabetes was induced to rats through intraperitoneal injection of streptozotocin solution (with a dose of 65 mg per kg of body weight). These rats were randomly divided into two groups, diabetic training and diabetic control; The training group performed MIIT for 6 weeks at an intensity equal to 55-75% of maximum speed. Data analysis was done through independent t-test in GraphPad Prism version 10 software. The significance level of the current research is P≤0.05.
Results: Total intracellular content of AMPKα1/2 protein did not show significant changes in the training group compared to the control group in the soleus skeletal muscle (P=0.96). In contrast, the phosphorylated intracellular content (P=0.0001) and the ratio of phosphorylated to total form (P=0.002) of AMPKα1/2 protein showed a significant increase.
Conclusion: MIIT increased the protein content of AMPKα1/2 in soleus muscle tissue of diabetic rats, and this could lead to increased energy production and consumption and improved glucose levels in diabetic subjects.
