Abstract

Background and aim: Cutaneous leishmaniasis (CL) is endemic in Iran and many other countries. The use of pentavalent antimony compounds for the treatment of this disease is associated with serious side effects. There have been reports of the effect of the extract of Berberis vulgaris root on leishmania major, both in vitro and in vivo. The goal of the present study was to investigate the effect of an alcoholic extract of this plant on experimental L. major ulcers in BALB/c mice. The experiment was conducted in 2006 by the Department of Parasitology, Kashan University of Medical sciences.

Materials and Methods: We used different concentrations of an alcoholic extract of Berberis vulgaris in the form of an ointment. Ninety BALB/c mice received a subcutaneous inoculation of 0.1 ml liquid-phase culture of infective L. major (MRHO/IR/75/ER standard variety) in the base of their tail. Ulcers appeared after 30-35 days and these were divided into 5 groups: 3 groups received treatment with 20%, 40%, and 80% concentrations and the remaining 2 groups served as controls. The extract was used applied topically, three times a day for 30 days. Each weak we measured the diameter of the lesions and the animals' weight. After preparation and staining, we determined the number of parasites in lesions through microscopic examinations. Results were recorded on paper and analyzed with the use of Student's t and Fisher's exact tests as well as analysis of variance and Tokey's post-hoc tests.

Results: With the 20% preparation: By the end of the treatment period, the mean diameter of the lesions had decreased, with complete healing in 5 mice (27.7%), (p< 0.001). By the time of the decrease in diameter, the mean weight of the animals had increased and the number of parasites in the lesions had declined (80%). Total elimination of the parasites was observed in 12 animals (p<0.001).
At a concentration of 40%: Mean ulcer diameter decreased, with complete healing in 2 mice (11.1%, p<0.001). By the time of the decrease in diameter, the mean weight of the mice had increased (p<0.05). The mean number of parasites in lesions decreased (64.3%), with total elimination in 9 animals (p< 0.05).

Conclusion: We showed that the extract of Berberis vulgaris is most effective at a concentration of 20%, causing greater reductions in both ulcer diameter and the number of parasites in the lesions compared to other preparations. Therefore, we suggest the use of a 20% extract for the treatment of human cases.

Abstract

Background and Aim: To determin the most sensitive tissues to carcinogenic effect of urethane and co-carcinogenic effect of sodium nitrite by investigation of histopathological variations in different tissues of balb/c mice.

Materials and Methods: An experimental study in 40 inbred balb/c mice aged 9-11 weeks was designed. The samples were classified into 4 groups: The first group was mice treated with 50 mg/l sodium nitrite in 0.9% NaCl instead drinking water SN&NaCl). The urethane group (U) was characterized by intraperitoneal injection of 3 times urethane (600 mg/ kg/day at 48 hour intervals). The third group was given 50 mg/l sodum nitrite in drinking water (U&SN) following the same intake of urethane as the second group, and the forth one was the control group. All mice were sacrificed after 20 weeks tissues were removed and examined for histopathological changes. Data analyzed by Fisher exact test.

Results: The formation of lung tumor was, significantly, increased in the urethane group as compared with the control group & (NS+Nacl) groups ( respectively P<.004 & P<.02) and in (U+NS) group as compared with the control group & (NS+Nacl) groups ( respectively P<.003 & P<.02). In females mice also The formation of lung tumor was, significantly, increased in the urethane group as compared with the control group & (NS+NaCl) groups (P<.05) and in the (U+NS) group was significantly higher than those in (C) & (NS+NaCl) groups (P<.009). The formation of lung adenocarcinomas was, significantly, increased in the (U+NS) group as compaired with the (C) & (NS+Nacl) groups (P<.003). In females mice also the formation of lung adenocarcinomas was, significantly, increased in the (U+NS) group as compaired with the (C) & (NS+Nacl) groups (P<.009) and (U) group (p<.05).The frequencies of stomach and testis hyperplasia and metaplasia in different groups did not show any significant differences. There was no histopathological variation in other tissues.

Conclusion: Our results showed the tumorigenic effect of urethane in lung tissue. Sodium nitrite increases tumorogenic effect of urethan. Sodium nitrite increases carcinogenic effect of urethan (This result is the first study that suggest sodium nitrite increases carcinogenic effect of urethan).Urethan increases co-carcinogenic effect of sodium nitrite.

Abstract

Background and Aim: Due to importance of employing native and traditional medicinal plants with good efficacy against malaria parasite, an ethanolic extract of Otostegia persica, was tested on Plasmodium berghei in sourian mice and a comparison was made with the effect of chloroquine on the same parasites.

Materials and Methods: In this Study 80 Sourian mice were divided in-to 8 groups , each consists ten animals. The first 7 groups were infected with P.berghei and the last group was used as control. The first seven groups were given chloroquine, Otostegia persica . at four different concentration (20,100 ,300,450mg /kg) and placebo.respectively and the seventh group did not receive any treatment. The evaluation was done by Rane test.In each group the level of parasitemia was determined on day 4 & 7 , and compard with values from day (just before treatment) in order to record the decline parasitemia in treated groups. Result were anylased using SPSS software and T-test.

Results:The result indicated that 450 mg/kg concentration Otostegia persica significantly decreased P.berghei parasitaemia in the infected mice (P<0.05), but not as sharp as chloroquine.

Conclusion:We can concluded that the effectiveness O.persica as P.berghei in 450 mg/kg concentration can be considered, for futures studies.

Abstract

Background and Aim: Medicinal plants provide an excellent source of drugs and new drug combinations, including the basis for the development of drug resistance to common anti-malarial treatments. In this study we investigated and compared the effect of an alcoholic extract of saffron and its constituents with that of chloroquine on malaria in mice.

Materials and Methods: A total of 65 male mice similar as regards gender, weight and age were randomly divided into 13 groups of 5 each. Eleven of the groups were infected with Plasmodium berghei and treated with saffron extract and its constituents and chloroquine based on the proposed method of Ryley and Petrs. Upon detecting the parasite in peripheral blood of the infected mice, they were treated with an aqueous, alcoholic or ethyl acetate solution at a dose of 350, 700 or 1050 mg/kg body weight (BW), chloroquine at a dose of 20mg/kg BW, and an iron chelator (50mg/kg BW). The solutions with the most effective concentrations were determined. Oral treatment at every stage lasted for up to 4 days. Parasite reduction in the groups treated with the extract was determined on days 4 and 7 and compared with that in the control, placebo and chloroquine groups. The Group 13 mice had no parasite injection and received no drug and were kept in the animal house only to control accidental death.

Results: Data analysis (t-test; SPSS) showed the most effective concentrations to be 700mg/kg ethyl acetate solution and 350 and 700mg/kg saffron extract, causing a significant reduction of parasitemia in the infected mice on day 7 (p<0.05).

Conclusion: The findings of this study indicate the potential effect of the alcoholic extract of saffron on Plasmodium berghei and, therefore, should receive proper attention.
